Ethyl Glucuronide Determination in Hair as an Indicator of Chronic Alcohol Abuse: a Fully Validated Method by Gc-ei-ms/ms

چکیده
Hair analysis is a powerful tool for the detection of chronic or past drug ingestion and in recent years it has also been exploited for chronic alcohol abuse. Ethyl glucuronide (EtG) determination in hair has been increasingly employed for diagnosing chronic excessive drinking in both clinical and forensic areas. This work describes a simple fully validated method for the determination of EtG in hair using an Agilent 7890B gas chromatograph coupled to an Agilent 7000C triple quadrupole mass spectrometer detector with an electron impact ion source (GC-EI-MS/MS). N-Methyl-N-(trimethylsilyl) trifluoroacetamide (MSTFA) was used as the derivatizing agent and dimethylformamide (DMF) as the solvent enhancer. Sample decontamination and treatment were performed according to Hastedt et al. (2012). Linearity of the assay was tested between 2.5 – 100 pg/mg. The sensitivity of the method, expressed as limit of quantitation (LOQ), was 2.5 pg/mg. The method has been applied to a number of real samples, with analytical results in the range of < LOQ to 39 pg/mg. Market Forensics and Toxicology Author(s) Jennifer P. Reply to Crunelle et al. about the Article ‘A Comparison Between Serum Carbohydrate-Deficient Transferrin and Hair Ethyl Glucuronide in Detecting Chronic Alcohol Consumption in Routine’.
AIMS In heavy alcohol consumption laboratory tests represent an objective evidence. In this study we compared older and newer biomarkers in blood and in hair. METHODS Carbohydrate-deficient transferrin (CDT), ethyl glucuronide (EtG), AST, ALT, GGT, MCV were measured in a large sample (n = 562). All people declared no alcohol consumption within the last 3 months. Serum CDT was measured by the ...
متن کاملA SPME-GC/MS procedure for the determination of fatty acid ethyl esters in hair for confirmation of abstinence test results.
Fatty acid ethyl esters (FAEE), direct metabolites of ethanol, are suitable alcohol markers that can be detected in different tissues. The determination of FAEE in hair can help to evaluate social and excessive alcohol consumption. Due to the presence of FAEE in the hair of teetotalers, proving alcohol abstinence seems to be impossible. To verify these results, an solid phase micro extraction-g...
متن کاملDetection of the Direct Alcohol Biomarker Ethyl Glucuronide (EtG) in Hair An Annotated Bibliography
Ethyl glucuronide (EtG) is considered to be a promising candidate marker of alcohol consumption, but exhibits a short window of detection in blood or urine. Keratinized tissues are known to retain foreign substances and to provide a greater retrospective window of detection than body fluids. Therefore, post-mortem hair, skin swabs, and stratum corneum samples were collected from four subjects w...
متن کاملQuantitation of ethyl glucuronide in serum & urine by gas chromatography - mass spectrometry
BACKGROUND & OBJECTIVES Alcohol misuse has now become a serious public health problem and early intervention is important in minimizing the harm. Biochemical markers of recent and high levels of alcohol consumption can play an important role in providing feedback regarding the health consequences of alcohol misuse. Existing markers are not sensitive to recent consumption and in detecting early ...
متن کاملGas chromatography tandem mass spectrometry for biomarkers of alcohol abuse in human hair.
We present the development of a new, faster analytical method for the determination of fatty acid ethyl esters (FAEEs) in human hair to detect potential alcohol abuse. FAEEs have been established as metabolites of ethanol consumption in humans and are embedded in the hair follicles during hair growth. The developed method has a total analysis time-including washing, extraction, concentration, s...
